Wissenschaftliche Hintergründe findet man in diesem Paper.

    Abstract

„Dark Therapy“, in which complete darkness is used as a mood stabilizer in bipolar disorder, roughly the converse of light therapy for depression, has support in several preliminary studies. Although data are limited, darkness itself appears to organize and stabilize circadian rhythms. Yet insuring complete darkness from 6 p.m. to 8 a.m. the following morning, as used in several studies thus far, is highly impractical and not accepted by patients. However, recent data on the physiology of human circadian rhythm suggests that „virtual darkness“ may be achievable by blocking blue wavelengths of light. A recently discovered retinal photoreceptor, whose fibers connect only to the biological clock region of the hypothalamus, has been shown to respond only to a narrow band of wavelengths around 450 nm. Amber-tinted safety glasses, which block transmission of these wavelengths, have already been shown to preserve normal nocturnal melatonin levels in a light environment which otherwise completely suppresses melatonin production. Therefore it may be possible to influence human circadian rhythms by using these lenses at night to blunt the impact of electrical light, particularly the blue light of ubiquitous television screens, by creating a „virtual darkness“. One way to investigate this would be to provide the lenses to patients with severe sleep disturbance of probable circadian origin. A preliminary case series herein demonstrates that some patients with bipolar disorder experience reduced sleep-onset latency with this approach, suggesting a circadian effect. If amber lenses can effectively simulate darkness, a broad range of conditions might respond to this inexpensive therapeutic tool: common forms of insomnia; sleep deprivation in nursing mothers; circadian rhythm disruption in shift workers; and perhaps even rapid cycling bipolar disorder, a difficult- to -treat variation of a common illness.

In this context, it is tempting to speculate that if CLOCK/BMAL1 acts as kamikaze TFs, as previously suggested46, transcription repression by homeodomain TFs may represent a mechanism to buffer CLOCK/BMAL1 high transcriptional bursts. In this framework, repression via the BMAL1 TAD could offer an efficient and simple mechanism for amplitude regulation and environmental entrainment. https://www.nature.com/articles/s41556-026-02041-4

Melatonin, ROR-α and circadian rhythm in liver

    Abstract

The liver is the largest internal organ. Several critical functions are attributed to the liver which include metabolism, synthesis of serum proteins, excretion, detoxification, and various physiological processes essential for maintaining body homeostasis. Its unique regenerative capacity helps the liver to restore itself fully after injury. This process involves all hepatocytes with or without the involvement of stem cells. The function of the liver is known to be regulated by circadian rhythm, which includes feeding-fasting cycles and the maintenance of the suprachiasmatic nucleus (SCN) that regulates as a master clock. The normal functioning of the liver is critical to the overall maintenance of homeostasis as it serves as a peripheral clock, suggesting a potential link between the SCN and liver. Aberrations in these circadian rhythms have been linked to various chronic hepatic diseases such as metabolic dysfunction-associated steatotic liver disease (MASLD), which can lead to Hepatocellular carcinoma (HCC). This mini review explores the significance of circadian rhythm in liver function, with a focus on the role of melatonin and nuclear receptors such as Retinoic acid receptor-related orphan receptor-alpha (RORα), which is a known melatonin receptor critical to sustaining these rhythms that can influence biological functions, including immune system functioning, cell growth, and differentiation. Further, RORα is identified as one of the key regulators of inflammation and acts as a potential tumor suppressor, particularly in the context of HCC. This review explores the interplay between RORα, melatonin, and circadian rhythm and discusses the underpinnings that offer insights into the role of circadian rhythm disruption in HCC development and novel therapeutic strategies targeting circadian rhythm modulations to mitigate HCC.

While our observational data cannot establish causality, previous experimental models suggest that the association between CircS and incident CLD might involve dysregulation of the “clock–lung axis”. The molecular clockwork orchestrates important pulmonary defense mechanisms, including the resolution of airway inflammation, the secretion of mucus, and redox balance3.

First, circadian misalignment decreases the expression of nuclear factor erythroid 2-related factor 2 (Nrf2), the master regulator of antioxidant responses3. In the CircS state, the rhythmic activation of Nrf2 is weakened, increasing the susceptibility of lung tissue to oxidative stress caused by environmental factors29.

Second, systemic metabolic stress promotes chronic low-grade inflammation (“meta-inflammation”)30. The increases in visceral adiposity and insulin resistance drive the secretion of proinflammatory cytokines (IL-6 and TNF-α), which can enter the pulmonary circulation, priming the lungs for exaggerated inflammatory responses and tissue remodeling31,32. However, because these specific cardiometabolic traits are embedded within our composite CircS definition, our cohort data preclude formal mediation analyses to statistically isolate this pathway. Therefore, this meta-inflammation mechanism remains a plausible hypothesis in our study context.

A key finding of our study is the stronger effect size observed in the ELSA cohort than in the CHARLS cohort. This difference may be partially explained by the synergistic effect of clustered risk factors. The ELSA cohort exhibited a significantly higher baseline BMI and greater prevalence of alcohol consumption. Alcohol is a potent chronodisruptor that uncouples the central pacemaker in the suprachiasmatic nucleus from the peripheral clocks in the liver and lung33,34. When CircS compromises baseline circadian integrity, concurrent lifestyle stressors such as alcohol consumption or severe obesity may further overwhelm the residual compensatory mechanisms of the lungs35,36. This view is supported by our subgroup analysis involving the ELSA cohort, where the risk was disproportionately concentrated among individuals who consumed alcohol, suggesting that lifestyle stressors can synergistically amplify the pathogenicity of circadian misalignment.
